western blot using anti-COI1 antibodies
Samples are 10-day old Arabidopsis thaliana seedlings of Col-0 and coi1-1 mutant (premature stop codon at W467* vs a full length protein of 592 aa), roots from 10-day old seedlings, either mock treated or treated with 20 uM MeJA. 20 ug of protein were separated on 4-12% NuPAGE gel and transferred onto PVDF membrane. The membrane was blocked with 5% skim milk dissolved in TBST. Primary antibody was added at a dilution of 1:1000 and incubated overnight at 4°C. The membrane was then washed 3X15 min with TBST at RT. Secondary antibody was added at a dilution of 1:10 000 (AS09 602, Agrisera) and incubated at RT for 1 hr, followed by washes for 3X15 min with TBST. The detection was performed using the ECL kit (SuperSignal West Pico from Thermo scientific).
