Kelch repeat-containing protein (KrP)/Galactose oxidase is a protein associated with iron acquisition and metal homeostasis which is a marker for iron defficiency sue to the fact that the transcript levels are stably regulated under iron defficiency conditions. It has been insinuated that to be a target of of bHLH39, bHLH100 and bHLH101 transcription factors. The gene localized at locus At3g07720 is a supposed ancestor of NSP (nitrile-specifier proteins), NSP1, NSP2, and NSP5, which unlike its descendants does not show nitrile-specifier activity.

Application example
Western blot with anti-KrP antibody

XX µg of total protein from Arabidopsis thaliana Col-O root or shoot grown on plates with (+) or without iron (-) extracted and denatured with 2X Laemmli buffer (write exact buffer components) at 95°C for 5 min followed by 5 min on ice. Samples were separated on XX% Mini-PROTEAN TGX Stain-Free gel and blotted XX min to nitrocelulose membrane using semi-dry/wet transfer. Blots were blocked with 5% milk powder dissolved in TBST for XX min at room temperature (RT)/ in a cold room with agitation. Blot was incubated in the primary antibody at a dilution of 1: 2000 for XX min at RT with agitation. The antibody solution was decanted and the blot was rinsed briefly twice, then washed XX times for XX min in TBST buffer at RT with agitation. Blot was incubated in secondary antibody (anti-rabbit IgG horse radish peroxidase conjugated, from ) diluted to 1:4000 in for XX min at RT with agitation. The blot was washed as above and developed for XX min with (EXACT NAME OF REAGENT AND SUPPLIER) using … detection system (name)/X-ray film. Exposure time was XX seconds.
