Rubisco catalyzes the rate-limiting step of CO2 fixation in photosynthesis.  This enzyme contains two subunits, each present in eight copies. In plants and green algae, 55-kD large subunit is coded by the chloroplast rbcL gene, and the 15-kD small subunit is coded by a family of nuclear RbcS genes.

Antigen used to elicit this antibody has been prepared in a following way:
1) intact chloropalsts from pea wee purified using Percoll density gradients
2) chloroplasts were hypotonically lysed and stroma was cleared of membranes by centrifugation
3) Rubisco Holoenzyme (L8S8) was isolated by sucrose density centrifugation
4) L8S8 complex was disrupted by pH shock and the L8 oligomer was precipitated
5) mature, folded Small subunit was further isolated by DEAE chromatography
6) It was considered pure by SDS-PAGE
