Arabidopsis thaliana leaves treated or not with Pseudomonas syringae containing the avirulence gene avrB: extracted with Tris-HCl 50mM pH 7.8, 0.1 mM EDTA, Triton X-100 0.2% and denatured with SDS 2% and DTT 10mM at 95ºC for 5 min. were separated on 12 % SDS-PAGE and blotted 1h to PVDF using semi-dry (Bio-Rad). Blots were blocked with 3% milk in TBS-T 1% O/N at 4ºC with agitation. Blot was incubated in the primary antibody at a dilution of 1: 5000 for 1h at RT with agitation. The antibody solution was decanted and the blot was rinsed, then washed 3 times for 10 min in TBS-T at RT with agitation. Blot was incubated in secondary antibody (anti-rabbit IgG horse radish peroxidase conjugated, from Agrisera) diluted to 1: 25 000 in TBS-T for 1h at RT with agitation. The blot was washed as above and developed for 5min with ECL Plus (Amersham). Exposure time was 45 seconds.
