Background

Background Agrisera Non Mammalian Protein Membrane Blocker is a blot friendly formulation for optimal blocking effect.

Recommendation for use:
  1. Following protein transfer to PVDF or nitrocellulose membrane, block the membrane with AgriBlock Non mammalian Protein Membrane Blocker, equilibrated to room temperature before use, for 1 hour at room temperature or overnight at 4ºC.
  2. Wash the membrane with TBST or PSBT three times per wash for 5 minutes, each with agitation on a shaker at room temperature.
  3. Perform further steps following western blot protocol for a given primary antibody:
  4. Add primary antibody of choice, at the appropriate optimized dilution using the recommended antibody diluent and incubate either 1h at room temperature or overnight at 4°C with agitation on a shaker.
  5. Wash the membrane with TBST of PBST, according to your protocol, with agitation on a shaker at room temperature.
  6. Add a matching secondary antibody at the appropriate optimized dilution and incubate for 1 hour with agitation on a shaker at room temperature.
  7. Wash the membrane with TBST of PBST, according to your protocol, with agitation on a shaker at room temperature.
  8. Proceed with visualization of reaction, depending upon the system which is applied, e.g. chemiluminescent (HRP-conjugated antibodies), chromogenic (ALP-conjugated antibodies) or fluorescent (for example DyLight®).
