Background COI1 (Coronate insensitive 1) is an esential component of jasmonate receptor. Required for jasmonate-regulated plant fertility and defense processes, and for coronatine and/or other elicitors perceptions/responses. Alternative names: F-box/LRR-repeat protein 2, AtCOI1, AtFBL2.
Immunogen Purified, recombinant COI1 protein, corresponding to amino acids: 350-592 of Arabidopsis thaliana COI1, overexpressed in E.coli. UniProt: O04197, TAIR: AT2G39940.

Application example

western blot using anti-COI1 monoclonal antibody

Total protein from Arabidopsis thaliana wild type (2) and knock-out mutant (1), extracted with protein extraction reagent and denatured at 95°C for 10 minutes were separated on 15% SDS-PAGE and blotted 1h to PVDF using wet transfer. Blots were blocked with 1×TBST+5% Nonfat dry milk for 1h at 37°C with agitation. Blot was incubated in the primary antibody at a dilution of 1:1000 in 1×TBST+5% Nonfat dry milk for 1h at RT with agitation. The antibody solution was decanted and the blot was rinsed briefly twice, then washed once for 15 min and 3 times for 5 min in TBS-T at RT with agitation. Blot was incubated in secondary antibody (anti-rabbit IgG horse radish peroxidase conjugated) diluted to 1:15 000 in 1x TBST + 5% Nonfat dry milk for 2h at RT with agitation. The blot was washed as above and developed for 5 min with ECL according to the manufacturer's instructions. Exposure time was 10-20 seconds.
