DCL1 (EC=3.1.26) is a ribonuclease (RNase) III involved in RNA-mediated post-transcriptional gene silencing (PTGS). Functions in the microRNAs (miRNAs) biogenesis pathway by cleaving primary miRNAs (pri-miRNAs) and precursor miRNAs (pre-miRNAs).

Application example
western blot using anti-DCL1 antibodies
Proteins from nuclear fraction and cytoplasmic fraction of Arabdiospsis thaliana were separated on  7 % SDS-PAGE using semi-dry transfer and blotted 1h to PVDF. Blots were blocked 2,5% milk in TBS-T  for over night at 4 degrees with agitation. Blot was incubated in the primary antibody (1:500; anti-DCL1, AS12 2102, lot 1508 (N) )) in PBS-T for 1h at RT with agitation. The antibody solution was decanted and the blot was washed 3 times for 15 min in PBS-T at RT with agitation. Blot was incubated in secondary antibody ( 1:10000; anti-rabbit IgG horse radish peroxidase conjugated, from Agrisera cat no. AS09 607) for 1h at RT with agitation. The blot was washed as above and developed for 5 min with ECL according to the manufacturer's instructions. Exposure time was 360 seconds
