Western blot using anti-V ATPase subunit A and B antibodies

20 µg of total proteins from Dunaliella tertiolecta (2)Dunaliella bardawil (3)Chlamydomonas reinhardtii (4) Arabisposis thaliana (6), Triticum aestivum (8) were extracted with 50mM HEPES buffer+PI and denatured at 70ºC for 5 min. Proteins were separated on 10% SDS-PAGE and blotted 1.5h nitrocellulose membrane using semi-dry transfer. Blot was blocked with 3% non-fat milk in PBS-T O.N at 4ºC. Blot was incubated in the primary ab at 1:2000 dilution for 1h at RT with agitation. The ab solution was decanted and the blot was washed 3 times for 10 min in TBST at RT with agitation. Blot was incubated with secondary ab (goat anti mouse IgG alkaline phosphatase conjugated) in 1:8000 dilution for 1.5h at RT with agitation. Blot was washed as above and developed for 10 min with NBT and BCIP solution.
