The blotting showed two bands at approx 30 kDa  and 12 kDa. A Nickel column-purified recombinant protein,  representing a cytoplasmic loop region of the ZIP7 protein,  was run on the gel. It was expressed in E. coli with a  HIS tag, and is 81 amino acids long, which equates to  the ~12-15 kDa band seen in the elution fractions of the western blot. The immunogens used were synthetic peptides  targeted to two regions within this cytoplasmic loop.
