Crude extract of Saccharomyces cerevisiae was separated on a 12.5 % SDS-PAGE and blotted to a PVDF membrane using wet transfer, following blocking with 5 % non-fat milk for 1 h/RT. Primary antibody was incubated at 1: 1000 for 1h/RT, followed by washes and incubation with a secondary goat anti-rabbit IgG HRP conjugated antibodies, used at 1: 10 000 1h/RT. Reaction was developed using chemiluminescence following manufacture's recommendations.
Samples: Recombinant Rad51 protein of Saccharomyces cerevisiae, analyzed by SDS-PAGE and visualized by Coomasie stain (1), 10 ng of recombinant Rad51 protein of Saccharomyces cerevisiae (2), crude extract of Saccharomyces cerevisiae strain BY4741 (3) were separated on a 12.5 % SDS-PAGE and blotted to a membrane using wet transfer. Primary antibody was incubated at 1: 1000, followed by washes and incubation with a secondary goat anti-rabbit IgG HRP conjugated antibodies, used at 1: 10 000 1h/RT. Background: S. cerevisiae Rad 51 protein (400 aa, 43 kDa) is a functional and structural homolog of E.coli RecA and human Rad51 proteins and plays a central role in DNA homologous recombination and recombination repair by promoting homologous DNA strand exchange reaction. Dmcl, Rad55, Rad57 are paralogs of Rad51 and they form complex with Rad51 and Rad52 in mediating recombination processes. - Product Citations
