  • Western blot using anti-Myc antibodies on nuclear cell extract from A. taliana

    20 µL of 150 µg/mL protein extract from nuclear fraction of A. thaliana dry seeds using the commercial buffer from BIORAD, laemmli 4x ref 1610747. Extracts were denatured for 5 min at 90°C and subjected to electrophoresis on BIORAD gradient gel (4-20%), mini-PROTEAN TGX gel (ref 4561093) at room temperature. The transfer was done for 7 minutes with the BIORAD semi-dry trans-blot turbo kit on nitrocellulose with the pore size of 0.2 µm. The blot was blocked with 5% of nonfat milk in PBST for 1h/RT. The blot was incubated with primary antibody at the dilution of 1:1000 ON/4°C with agitation, in 3% milk in PBST. The blot was washed 3 times for 5 minutes in PBST, under agitation at RT. The secondary antibody was diluted to 1:5000 dilution, and incubated for 2 h/RT with agitation in 3% nonfat milk in PBST. The blot was developed using chemiluminescent detection reagnet according to manufacture's recommendations.
