western blot using anti-PsbN antibodies

5 µg of membrane proteins from Nicotiana tabacum WT and Δ psbN mutant isolated with homogenization buffer (50 mM Tris HCl pH 8.0, 10 mM EDTA, 2 mM EGTA, 10 mM DTT ) were separated on 15% SDS-PAGE and blotted 1h to PVDF using semi-dry transfer. Blots were blocked with 3% BSA for 1h at room temperature (RT) with agitation. Blot was incubated in the primary antibody at a dilution of 1:1000 for 12h at 4°C with agitation. The antibody solution was decanted and the blot was rinsed briefly twice, then washed once for 15 min and 3 times for 5 min in TBS-T at RT with agitation. Blot was incubated in secondary antibody (goat anti-rabbit IgG horse radish peroxidase conjugated, AS09 602, Agrisera) diluted to 1:25 000 in BSA for 1h at RT with agitation. The blot was washed as above and developed for 5 min with ECL according to the manufacturer's instructions. Exposure time was 53 seconds.

The PsbN protein is chloroplast-encoded, low molecular weight protein annotated as a photosystem II subunit however it seems that it is not a constituent subunit of PSII but is required for PSII repair from photoinhibition.
