    western blot using hen anti-Cu Zn SOD antibody

    25 µg of total protein from Olea europaea pollen were separated on 12% acrylamide gels and blotted onto PVDF. Membranes were blocked 1h with 2% low-fat milk powder in TBS-T (0.1% TWEEN 20) and probed with anti-Cu/Zn SOD (AS10 652, 1:1500, overnight, 4º C) and secondary antibody anti-chicken IgG conjugated to Alexa 488 (1:2000, 1 h) (Molecular Probes, Invitrogen). Signal was detected with a PharosSX fluorescence scanner (Bio-Rad).

    The antibody recognizes a protein of c.a. 16 kDa. and another band likely corresponding to the protein dimer.

    We have used PC antibody with protein extrac from Hordeum vulgare.We got a special bandLoad per well: 15g total Protein, 14% SDS-GelMembrane: NC von Roth, HP40.1Blocken: 5% milk in TBST, 2h, RTCU/ZnSOD ( AS10 652 ): 1:1000 in 2% milk, 1 x TBST, over night, 4CSecondary antibody : Rabbit anti goat IgG AS 09605 1:50000 in 2% milk / 1 x TBST 1h RT Developing reagent: ECL ( GE Healthcare )1:1:2Exposure time: 1sec.

    I used this antybody for Cu/ZnSOD immunodetection analysis in the leaves of three grasses: Echinochloa crus-galli, Digitaria sanguinalis and Zea mays plants (all NADP-ME subtype). For each treatment 15 g of total proteins extract from the leaves were loaded on SDS-PAGE (12% acrylamide) and immunodetected with anti-Cu/ZnSOD (Agrisera). The highest signal was observed in Echinochloa crus-galli, Digitaria sanguinalis, and the weakest signal was noted in Zea mays in different irradiation of the light.
