Dot blot: using anti-m6A mouse monoclonal antibody with DNA oligonucleotide containing different m6A modified bases (in a range from 4 to 100 pmol) and a negative control which were spotted on a membrane (Amersham Hybond-N+). The membrane was incubated with the mouse anti-m6A monoclonal antibody (dilution 1:500) in TBS-T with a 10 % non-fat-dry milk.

Background
m6A (N6-methyladenosine) is a modified base, abundant in mRNA in most eukaryotes and found in tRNA’s, rRNA’s, snRNA’s and in long non-coding RNA’s. Adenosine methylation is catalyzed by m6A methyltransferase, a large protein complex which has a preference for the consensus sequence GGACU.
