Oftentimes TBS-T is a buffer of choice applied in many Western blot protocols for all antibody incubation steps. 

However, as you can see in this example, changing to PBS-T has also changed a pattern of detected bands. Analyzed samples (Arabidopsis thaliana), protein load/well, primary, secondary antibodies and detection reagent were kept the same.

Antibody incubation buffer has been changed from TBS-T (left panel) to PBS-T (right panel).

If you aim to detect phosphorylation, TBS-T containing blocking buffer is recommended, as phospho-specific antibodies may bind to phosphate in PBS buffer, and this will lead to decrease in detected signal.
