Waste less of resources in the lab!

How to cite antibodies

Can listings a product numbers of primary and secondary antibody used in your research actually minimize waste of work and resources in the future?

More than half of the antibodies currently used in research cannot be properly identified, which makes it impossible to conduct a given experiment using the same set of antibodies. Companies often offer multiple antibodies to the same target protein (e.g. tags and protein modifications) and identifying a specific antibody used in a given publication is simply impossible.

There are no worldwide standard guidelines on how antibodies should be cited in publications and journals. Besides the product number for any primary antibody used in research, the secondary antibody should also be properly cited with product number. As primary and secondary antibodies in a successful experiment create a unique pair, such result may not be reproducible with another secondary antibody. Antibodies differ in their characteristics, and lots of time and money can be saved in the future if product numbers of both primary and secondary antibodies are listed in all publications.

You can also read about this topic in Citeab blog post: "Don’t forget about citing your secondaries!"

Common misconceptions about antibodies

Can the same protocol can be applied to all antibodies?

No, this is NOT the case.

Each protein-antibody pair may require slightly modified binding and detection conditions. The variations may for example be a membrane with another pore size and physical properties (nitrocellulose or PVDF) as well as different blockers, incubation times and primary antibody dilutions. Some antibodies may work better in TBS-T buffer, others with PBS-T.

These issues are disussed in Agrisera Western blot guide, which we recommend you to keep in your lab!

Why fractionate?

Advantages of fractionation

If no signal is detected in a total cell extract, it is worth fractionating, to obtain specific chloroplast, mitochondria or nuclei enriched fractions. This procedure will concentrate a target protein and lead to the detection of a desired band (for non-cytoplasmic proteins). This is shown in the example above - from no band in a total cell extract, to a specific band in photosystem I fraction.
This approach is especially important when working with proteins of low expression. Fractionation combined with using a specific organ, e.g. the root tip, or a leaf at a certain developmental stage, will greatly contribute to the successful identification of your protein of interest.

Examples of protocols to fractionate can be found here.

Is antibody always to blame?

Western blot using anti-BAK1 antibodies

In the example above, the same antibody was used (Anti-BAK1, AS12 1858) but the proteins were extracted with two different buffers.
Minor adjustments in the buffer components resulted in the improved detection of the target protein - from NO band to a band of correct MW.

If you are interested in more tricks to optimize your western blot, you are welcome to contact us or request a copy of the Agrisera Western Blot Guide.

Simultaneous detection with two primary antibodies

Two primary antibodies incubated on the blot at the same time - save time in the lab

Western blot using anti-PetA antibodies

Thylakoid membranes (10 µg of total chlorophyll) extracted freshly from Hordeum vulgare leaves with 100 mM HEPES-KOH (pH 7.5), 0.3 M sorbitol, 2 mM EDTA, and 1mM MgCl2 and denatured with a Laemmli buffer at 80°C for 5 min were separated on 12% SDS-PAGE and blotted 1 h to nitrocellulose (pore size of 0.2 um), using semi-dry transfer. Blot was blocked with 4% milk for 2 h/RT with agitation. Blot was incubated in the primary antibody at a dilution of 1:3000 (PC and PetA, simultaneous western blot detection for both antibodies at the same time) for 1 h/RT with agitation in PBS-T. The antibody solution was decanted and the blot was rinsed briefly, then washed 3 times for 5 min in PBS-T at RT with agitation. Blot was incubated in Agrisera matching secondary antibody (anti-rabbit IgG horse radish peroxidase conjugated) diluted to 1:25000 in for 1 h/RT with agitation. The blot was washed as above and developed for 5 min with chemiluminescent detection reagent according to manufactures recommendations. Exposure time was 30 seconds. Simultaneous western blot detection can be applied if MW of detected proteins differs in min. 20 kDa.

Agrisera on Botanikertagung 2019, International Plant Science Conference

Agrisera at Botanikertagung 2019

Between 15.09-19.09 Agrisera participated in Botanikertagung, International Plant Science Conference, which is a biannual meeting of German plant and algal scientists. This year it was held at Rostock University, Germany.

An Agrisera quiz, where the participants got to test their weight determining skills by predicting the weight of the Agrisera chemiluminescent reagents/secondary antibody set, attracted a lot of attention, and the winner actually provided the exact answer!

Our third educational poster, Regulation of photosynthesis was so popular that we ran out of it. Additional copies will be sent by post. Thank you to all who came by the Agrisera table and talked with us about western blot and antibody production service or shared their experience with antibody work. We will hopefully meet again in two years in Bonn!

How to cite the antibodies used in scientific publications?

Agrisera antibodies in scientific publications

As more than half of the antibodies used in research cannot be properly identified, it is absolutely crucial to start including information in the documentation of scientific research, to allow antibody identification by researchers who want to repeat your experiments. Some journals provide a guideline for antibody citation, but unfortunately most of them do not. This vital issue is most often completely overlooked.

If there are no recommendations from the journal where you intend to publish your work, citing an antibody can be done in the following way:

Antibody name, product number, (supplier name, country)

For example: Anti-RbcL, AS03 037 (Agrisera, Sweden)

We would like to encourage you to cite antibodies in a way that will allow their future identification. This is especially important for antibodies to tag proteins.

Good luck with your research!

Plant meeting calendar is on-line!

Plant Meeting Calendar

After many years of supporting hundreds of plant conferences, workshops and congresses the idea of a reliable and global Plant Science Event calendar was born. Agrisera contacted American Society of Plant Biology and our idea seem to hit the right timing. After discussions and brain storming the calendar started to take shape and since January 2019 all plant meetings, congresses, workshops and conferences can be signed up to the Global Plant Science Events Calendar. The aim is to cover most of worldwide meetings and provide a comprehensive listing. At the moment there are over 65 meetings registered already.

The calendar offers following functionality:
  • There are few ways to easy receive an overwiev on coming meetings
  • Using a simple tool, plant event can be easily included into the calendar
  • A mini calendar widget can be loaded to a conference/society website, to obtain a widget contact: [email protected]
  • There is an option to subscribe to the calendar
  • Simple search can be done using specific key words
News about the calendar are spread worldwide, to reach plant community and we hope that it is going to be a very useful tool to sign up a meeting or check what is coming!
