Protein Extration Buffer - 50 % off until end of 2020

Protein Extraction Buffer (PEB) is a universal extration buffer for the disruption and solubilisation of total protein from plant tissue (root, leaf) and algal cells. It contains anionic detergent LDS, which together with the recommended procedure, which is a combination of sonication and freeze/thaw cycles, has been shown to increase the amount of solublised and non-degraded proteins, compared to other methods.

The PEB product information sheet can be found here.

Check: Video Tutorial: Sample harvesting and extraction

Protocols to work with plant and algal protein extracts

Please note that some proteins may require specific extraction conditions, as they may degrade in light or during extraction. Therefore, protein extraction protocol has to be modified accordingly.

Secondary antibodies, blocking and detection reagents

Agrisera Reagents

Agrisera offers a range of universal reagents, required in every laboratory using Western blot, ELISA or immunolocalization techniques.

Secondary antibodies
Over 2000 validated, high-quality antibodies for immunoglobulins from different species:

Anti-Bovine, Anti-Cat, Anti-Chicken, Anti-Dog, Anti-Donkey, Anti-Goat, Anti-Guinea pig, Anti-Hamster, Anti-Human, Anti-Llama, Anti-Mouse, Anti-Rabbit, Anti-Rat, Anti-Sheep.

These are offered both conjugated and unconjugated, for luminescent, fluorescent and chromogenic detection in a wide range of applications, such as ELISA, Immunolocalization studies and Western blot.

Detection reagents
Agrisera chemiluminescent detection reagents are developed using unique chemistry with low pico to mid femtogram and extreme low femto detection levels. These reagents also offer increased stability, 24 month shelf life and superior lot-to-lot consistency at a very competitive price.

Agrisera BCIP/NBT ALP Substrate is designed to localize ALP labeled probes on Western, Northern, Southern and Dot blots, while Agrisera BCIP/NBT Plus ALP Substrate is designed for immunohistochemical, in situ hybridization and blotting techniques. These chromogenic reagents are ready-to-use, single component, give no to little background and contain non-toxic stabilizers.

Blocking reagents
For most immunoassays like IHC, ELISA or Western blot, blocking is a crucial step to perform. It blocks spaces that are not occupied by immobilized proteins, which prevents non-specific binding of the antibodies used in the assay.

What is the safest method to store antibodies?

Proper antibody storage is crucial for the success of future experiments.

When it comes to antibody storage the rule "one method for all antibodies" does not apply, and it is not safe to assume that -80°C is always the best storage condition. Let us explain why. Antibodies are proteins of different stability, and therefore some antibodies can lose activity when stored at -80°C or on dry ice. This is also true in regards to the shipment of antibodies. Based on our experience and published references, some proteins may be damaged by storage or shipment on dry ice: Murphy et al. (2013): Protein instability following transport or storage on dry ice

For custom produced antibodies, we recommend testing various conditions to find the most suitable one:

• -20°C or -80°C
• +4°C with preservatives like azide (0.02%) or merthiolate
• -20°C with glycerol at a final concentration of 10% or 50% (for affinity purified antibodies)
• -20°C with BSA at final concentration of 0.05-0.5% (for affinity purified antibodies)

More information on what to consider when storing different kinds of antibodies can be found here

A known problem is that affinity purified antibodies can precipitate when stored at 4°C. However, such antibodies may still maintain their activity towards a target protein.

For catalog antibodies, we highly recommend checking the product information sheet, which is always included with the product, and contains proper information about storage conditions. If you have further questions, you can request additional information by contacting us

Good luck with your research!

Most published antibody to most abundant protein on Earth - Rubisco

Rubisco - most abundant protein on Earth

Rubisco is an amazing protein, and also the most abundant protein on Earth. Agrisera has provided specific antibodies to this enzyme for over 17 years. These antibodies have been cited in thousands of publications and used on a very wide range of species. The Agrisera anti-RbcL form I antibodies are made to a very conserved, 7 amino acid long peptide, and is therefore categorized as a Global Antibody. The specific peptide used is present in Rubisco form I from higher plants, algae and cyanobactera:

Anti-RbcL I | Rubisco large subunit, form I antibodies (AS03 037) - provided at a 50 % discount in February.

Combined with the Rubisco protein standard (AS01 017S) and the Rubisco Western Blot quantitation kit (AS09 409), this antibody can be used for quantification of Rubisco using quantitative western blot method. A short video tutorial for this method can be found here

For photosynthetic bacteria, archea and dinoflagellates, Agrisera offers another antibody:

Anti-RbcL II | Rubisco large subunit, form II antibodies (AS15 2955) - provided at a 50 % discount in February.

This antibody can be combined with RbcL II quantification standard (AS15 2955S) and applied to quantify RbcL II.

Agrisera's Rubisco antibody collection also consists of other useful antibodies, like: RbcS (AS07 259A), Rubisco activase (AS10 700), Rubisco chaperones and other proteins. The whole collection can be found here

Good luck with your research!

One year of The Global Plant Events Calendar

Word Cloud for The Global Plant Events Calendar

We are celebrating the 1-year anniversary of the Global Plant Events Calendar!

The Global calendar project was initiated on January 2nd, 2019 as a mutual effort of Plantae and Agrisera to provide the plant community with a comprehensive list of worldwide events.

We hope that the calendar will provide a quick overview of plant meetings, conferences and congresses, to help you decide and plan which conference to attend. The calendar will also help promote a meeting, should you be organizing one. The calendar widget can easily be loaded on the conference website.

The Global Plant Events Calendar has already collected over 200 events! There are over 72 events registered for this year and even two events for 2021. We are looking forward to new conferences to be signed in and hope that we can cover most plant meetings taking place in the world.

Not familiar with the calendar yet? This short video will show you how to use it.

Agrisera Prize 2019 awarded at Umeć Plant Science Centre

Agrisera Prize 2019 at Umeå Plant Science Centre

Jan Karlsson received the UPSC Agrisera Prize 2019 to acknowledge his ceaseless technical support for the UPSC community at UPSC (Umeå Plant Science Centre). The announcement was done during the Christmas lunch, and the motivation from one of the nominations was as follows: "He is always there for the whole institute to help to solve all kind of technical, computational, mechanical or physical strength-requiring problems. He has spent the night at work to make sure that the backup power system will not fail when there was a planned overnight power interruption. He did a tremendous amount of work with Thomas Hiltonen for the moving of Plant Physiology Department during the moving out from old labs, the reconstruction and moving into the new lab. He supervises all growth chambers, shakers and cabinets, and he is the one, who we hope to find in his office when we need a solution for a challenging technical issue. People can always ask for his advice or help, independently from which department they belong to."

This year the prize was presented by Joanna Porankiewicz-Asplund from Agrisera and by the chair of the UPSC Board, Catherine Bellini. From left to right: Joanna Porankiewicz-Asplund (Agrisera), Catherine Bellini (Chair of the UPSC board), Jan Karlsson. Photo: Anne Honsel.

Common misconceptions about antibodies

I have purchased several antibodies from the same company, and they all worked on my species. Therefore the next one should work too!

Each antibody is made to a different protein and thus recognizes different epitopes. Therefore, even if many antibodies purchased from a given company worked well for your species, this does not guarantee that the next antibody will. There needs to be a match of the sequence the antibody is binding to in the sequence of the protein which is aimed to be detected. Based on our experience, at least a 70 % match in linear sequence needs to be confirmed. Many antibodies are made to short, 10 to 15 amino acids long peptides. Overall target protein conservation between species will not be relevant in such a case if there is no match in the exact sequence used to elicit the given antibody.

Common Misconceptions About Antibodies

Therefore, before purchasing an antibody which has not been used on a given species or application, conservation of the antigen used to elicit such antibody should be confirmed.

Send your question to: [email protected] and we will help to check sequence conservation and confirm your choice for the best outcome of your experiments.
