20 % off high quality blocking serum and negative controls

20 % off blocking (normal) serum and negative control

20 % off Agrisera blocking (normal) serum

Agrisera offers a wide range of high-quality blocking (normal) serum for the application as a blocking agent, or as a negative control for immunoassays. All serum is obtained from healthy animals. The following formats are available:

  • Serum
  • Total immunoglobulin fraction
  • Highly purified, total immunoglobulin fraction
From the following species: Bovine | Chicken | Donkey | Duck | Goat | Guinea pig | Hamster | Horse | Human | Llama | Mouse | Pig | Rabbit | Rat | Sheep

Serum is lipid-extracted, dialyzed against phosphate buffer, and provided as lyophilized powder without preservative in different amounts:

From 1-10 mg to 1 g and bulk.

For most immunoassays like IHC, ELISA or Western blot, blocking is a crucial step to perform. This procedure blocks spaces that are not occupied by immobilized proteins, which prevents non-specific binding of the antibodies used in the assay. If blocking is not performed, false signals or high background will occur.

There is no universal blocking agent, therefore empirical testing is necessary to determine which agent to use for a given assay.

If normal serum is used as a blocker, it is recommended to use serum derived from the same host species as the detection antibody, conjugated with the reporter enzyme like HRP (Horse radish peroxidase) or ALP (Alkaline phosphatase).

Example:
Primary antibody: Rabbit anti-target

Blocking serum: Normal Chicken Serum (usually used at 5 % v/v in PBS with Tween®20 detergent)

Secondary antibody: Chicken anti-Rabbit IgG-HRP conjugated

Note: Rabbit serum cannot be used for blocking in such a combination.

Agrisera antibody used to evaluate Nanoalgosomes

Microalgae can be used to produce a novel, membranous, biogenic nanomaterial - Nanoalgosomes.

Nanoalgosomes can be utilized as a new natural delivery system for a range of high-value microalgal substances, such as antioxidants, pigments, lipids, complex carbohydrates, bioactive biological molecules (e.g., proteins, miRNA, siRNA, mRNA, lncRNA, peptides) and/or synthetic drugs.

Agrisera anti-H+ATPase antibodies, a marker of the plasma membrane, were used to characterize Nanoalgosomes, which has been experimentally demonstrated in a recent publication by Adamo et al. "Nanoalgosomes: Introducing extracellular vesicles produced by microalgae", J Extracell Vesicles. 2021 Apr; 10(6): e12081.

15 % off on antibodies for epigenetic research

Epigenetic research antibody promotion

Agrisera Antibodies for epigenetic research

Gene expression is regulated by epigenetic modifications, which occur either on histones (associated with DNA), on chromatin, or directly on DNA. These modifications include:

  • Methylation of DNA (transcriptional silencing)
  • Histone methylation
  • Histone acetylation

The Agrisera anti-Epigenetic research antibodies include antibodies to the following modifications:

  • H3K4me1 and H3K27ac - these modifications mark enhancers, regions of the genome of cell-type specific activation of gene expression
  • H3K4me3 - modification found in the promoters and active genes
  • H3K9me3 - marks constitutively repressed heterochromatin and works together with DNA methylation
  • H3K27me3 - marks regions of the genome, which are transcriptionally inactive
  • H3K36me3 - modification linked to transcriptional elongation and therefore associated with gene expression
  • 5-mC - addition of a methyl group at the 5 position of cytosines is associated with gene silencing
  • 5-hmC - is a DNA modification and results from the enzymatic oxidation of 5-mC by the TET family of cytosine oxygenases. 5-hmC modification at a gene is associated with its expression level

Do you know where exactly Agrisera is located?

Agrisera Winter 2021

Did you know that Agrisera is located even further north than Anchorage, Alaska and 3000 km from the North Pole?

Yes, this is correct. The temperatures in the winter can drop below -20ºC (good for storing antibodies!) and there can be days when we will receive over 50 cm of snow. Like this winter. In spite of slightly extreme conditions, we continue to work, produce and purify antibodies. The shipment of our antibodies worldwide also continues as normal, to support research efforts in plant science and other areas.

We pride ourselves in being, as our customers describe it: fast, cooperative and efficient!

New epitope tag antibody (BirA (mutated/TurboID) ) for protein interaction studies

Proximity labelling is a laboratory technique which makes it possible to label proteins (in this case by biotinylation), proximal to protein of interest. This allows studying protein interactome networks in different types of cells and organs, as well as different developmental stages. The technique is achieved by a gene fusion in a living cell, between the protein of interest and BioID (BirA). The TurboID variant is a biotin ligase, with substantially higher activity, which provides higher temporal resolution and broader application in vivo. The labelling time is just 10 minutes in RT, compared to the over 18 hours of labelling time required for BioID.

Agrisera offers a new polyclonal antibody, prepared to a recombinant mutated BirA protein from E.coli, called TurboID.
anti-BirA (mutated/TurboID)

The proximity labelling technique is described in various publications, e.g.:
Zhang et al. (2020). TurboID-Based Proximity Labeling for In Planta Identification of Protein-Protein Interaction Networks.J Vis Exp. 2020 May 17;(159).doi: 10.3791/60728.

Agrisera anti-TAG antibody collection is growing

Antibodies to tags can be applied in cases when there are no dedicated antibodies against a given protein. Addition of a specific tag, like for example FLAG, His, HA, makes it possible to study target protein using specific anti-tag antibody applied in techniques like: immunofluorescence, immunoprecipiation or Western blot or purification using affinity chromatography.

The peptide sequence of a tag can be fused to N-terminus or C-terminus of a protein under investigation or inserted  within a protein. Some tags, like FLAG can be combined with other affinity tags: His, HA or myc. Historically, FLAG is a first epitope tag and the only epitope tag which is patented.

Tag sequences are very short and recognized by high affinity antibodies, produced in wide species range: goat, chicken, mouse, rabbit.

Some of available anti-tag antibodies are tag position sensitive and one has to take it into account, before a purchase of anti-tag antibody.

HSP70 ELISA kit discounted until end of 2020

HSP70 (Heat Shock Protein 70) is a monomeric protein which, under physiological conditions, functions as a molecular chaperone in an ATP-dependent manner, protecting cells against lethal damage induced by stress.

By means of a Agrisera's sandwich ELISA kit (AS17 4128) it is possible to quantitate cytoplasmic plant HSP70 concentration in plant extracts from Medicago sativa, Linum usitatissimum, Zea mays and other species. The plate is coated with anti-plant HSP70 capture antibody, and 40 samples can be analyzed in duplicate

Sensitivity: 0.18 ng/ml | Assay range: 1.563-100 ng/ml | Incubation time: 30 min

The kit is discounted until end of 2020.

Agrisera Loading Control Antibody Collection

Loading controls are used to normalize the levels of proteins detected using Western blotting, and will:

  • confirm that there is an equal loading across the gel
  • confirm that the protein transfer is even throughout the whole gel
  • allow qantification
  • can be requested by referees of scientific publications

Proteins of high levels of constitutive expression are usually recommended as loading controls. However, the choice of the appropriate house-keeping protein depends on the experiment in question, and thus needs to be confirmed. House-keeping proteins cannot be used for normalization if there is a suspicion that the experimental conditions are affecting their expression.

Agrisera's Collection of Antibodies to Loading Controls can be found here. Until end of 2020 they are offered at 15 % off.
